to assign the 3rd column as x and 2nd column for y:

plot 'xxx.yyy' using 3:2

see help using and help plot.

I don't know if I'm understanding exactly what you want to do, but to plot =
a
column as -y, then:

plot 'xxx.yyy' using 3:(-$2)

again, see help using and help plot.
