Pretty sure this won't work as the database file is always open so it won't be able to be updated. But give it a try might be able to work around things if you are creative.

Not true. The PortableApps.com platform can be installed onto any Cloud storage site, like OneDrive, Dropbox, etc., and Ditto is one of the apps specifically configured to work in this environment. I store it in my OneDrive and it works like a charm.

Since you can specify the path to the database file, I suppose it will simply work.

It works with oneDrive (using Ditto portable).
But oneDrive will always complain, that it can't sync the file Ditto.db
