I absolutely LOVE Portable Scribus.
It works like a dream - even the PDF output is perfect, unlike with the very expensive PageMaker and Distiller!
Two features that were in PageMaker 6.5 which I used to have, ages ago, which are not in this newer software, which I would love to see, are Autoflow, and moveable page guides that actually stay visible on the page once you place them...

I have no clue. It should be fine theoretically, if a profile has already been created. Keep in mind that any changes you make to any settings probably won't save.
You could test it out by setting the Portable Scribus directory to read-only, and see if/how it works.

Is it possible to run Portable Scribus from a non-writeable media (like CD, write-protected usb-stick) ?
Which settings do I have to adjust for that (to prevent Scribus to write back settings .....) ?.

Portable Scribus 1.3.3.11 has been released. Includes Scribus 1.3.3.11 and Ghostscript 8.61. Changes include a basic language changer and a Scribus launcher tweak.

Yes. The latest 1.3.3.11 version has a language changer.
Otherwise you can just edit the "Language=en" field to "Language=de" in scribus.ini.
