Multiple minor but useful improvements since the (rather too hasty) v3.0.0 release including:
- Significantly faster start-up time.
- The in-silico PCR checking function now warns if any primer pair is predicted to amplify more than one product.
- Fixes an issue where the in-silico PCR check would fail when targets existed for which no primers were designed.
- Improved visibility of progress messages in the main interface.
- Fixes an issue with incorrect numbering of exons at the gene level.
- Improved instructions.
AUTOPRIMER3
Automatically design primers to genes or coordinates.
This is the redesigned AutoPrimer3 that no longer requires a large local database and is no longer limited to human genes but instead can connect to any UCSC genome.
Versions are available for Mac OSX (tested on Yosemite), Windows (tested on Windows 7) and Linux (tested on Arch, Ubuntu 14.04 and CentOS 7). Only 64-bit operating systems are supported.
INSTALL
64 bit systems only
Linux: Download the AutoPrimer3-3.0.1-1.x86_64.rpm (for RPM-based distros - e.g. CentOS), autoprimer3-3.0.1.deb (for DEB based distros, e.g. Ubuntu) or AutoPrimer3-3.0.1-1-x86_64.pkg.tar.xz (Arch Linux) depending on your system and install via your package manager (e.g. sudo dpkg -i autoprimer3-3.0.0.deb) or, if your distribution supports it, by double-clicking the downloaded package.
Mac OS X: Download the AutoPrimer3-3.0.1_macOSX.dmg disk image. Double click to open the downloaded file and drag the AutoPrimer3 application into your Applications folder where it can be opened by double clicking. On recent versions of Mac OS X you will need to access the 'Security & Privacy' settings in System Preferences to allow this app to be opened.
Windows: Download the AutoPrimer3-3.0.1_installer.exe installer application. Double click the downloaded file and follow the instructions to install. This will also launch AutoPrimer3 once installed. To launch in future it should be accessible from the Start menu->All Programs->AutoPrimer3. AutoPrimer3 will be installed to C:\Users[Your Username]\AppData\Local\AutoPrimer3\AutoPrimer3.exe.
UNINSTALL
Linux: Uninstall via your package manager (e.g. sudo apt-get remove autoprimer3).
Mac OS X: Drag the AutoPrimer3 application from your Applications folder to the recycle bin.
Windows 7: AutoPrimer3 should be installed to 'C:\Users[Your Username]\AppData\Local\AutoPrimer3\AutoPrimer3.exe'. To uninstall double-click the 'unins000.exe' file in this location. You can also access this location by right-clicking the AutoPrimer3 application from the Start menu, selecting 'Properties' and clicking the 'Open File Location' button in the resulting window.
AutoPrimer3 also stores a small amount of data (~2MB) in a folder called .AutoPrimer3 within your home folder. Manually delete this folder if you want to remove all traces of AutoPrimer3 from your machine.
