I'm reckoning there is a corruption or similar in your virtuser map. Check the actual configuration file, to make sure it 'looks' right. The first thing I do in these situations is check the log for the server daemon in question...perhaps do a service restart of Sendmail and see if it complains about something wrong with one of your configuration files.
