The inittex.ini file should (or always used to be) generated after
installation. I've spotted that that hasn't happened with a couple of
recent installations, and I've just submitted it as an issue on github.
You can generate it by running "gle.exe -mkinittex" form a command prompt
(if gle.exe isn't in your path, you'll have to use the full path or run it
from the GLE bin directory). (You can define new symbols in the init.tex
file and then recreate the inittex.ini file to have them available for use.)
