There are fonts which have this as a ``feature'' such as David Lemon's Copal, or, Rosewood and some of the other Adobe Wood Type fonts --- it's typically done by using 1 character per desired colour, so for a black font w/ white knockouts one would design a pair of characters, one a zero-width, solid one for the white (which would go first), then a second cut out one for the black.
