Webmin's Samba module can support any backend that the pdbedit command
can handle. When you view and edit users, it first tries to use the pdbedit
command (which comes with Samba), and falls back to directly edit smbpasswd
if pdbedit is not installed.

From my understanding, pdbedit looks and smb.conf and uses whatever backend
(such as LDAP or tdbsam) is configured there. If this is not working for you,
try clicking on the Module Config link in the Samba module, and making sure
that the 'Full path to pdbedit' is set correctly.
