Samples sure sound good, but they tend to be a bit static, something that a well thought out synthesis algorithm can overcome. If you don't use the softsynths directly in MusE, but instead go through the ALSA sequencer both timidity and iiwusynth are supported on par. The only problem might be, that most soundcards don't support two programs to share one device, so you're left with either MusE or timidiy/iiwusynth using the soundcard. I have this problem using PD with MusE, which is why I try to keep everything regarding audio inside of PD: synthesis, sampleplaying (iiwu~), ...
