Nest-o-Patch 2.1.10.1

Command "Set axis position and grid distance" added for the Trace. It allows to set convenient rounded grid distance and axis position. Can be useful if for example you want to use Nest-o-patch screenshots in a presentation. Warning: these settings will be automatically changed if you move or rescale your trace, so, set them directly before taking a screenshot.

Nest-o-Patch 2.1.9.4

Minor bug fix release. In the ver. 2.1.9.3, Mean and CV for opening times were not shown in the "Single channle kinetics" Tab. Now this is fixed.

Algorithm of multigaussian fit of the current histogam was refined.

Complete support of files recorded with PatchMaster 2x90.4. Files can be opened and all information is correctly displayed. ReadPMaster and ReadPulse ver. 4.4 are necessary (and included in the package).

Special bug-fix release. Bug in ReadPMaster.dll which prevented PatchMaster file opening in rare case when PatchMaster incorrectly saves series resistance, was fixed.

No changes in Nest-o-Patch itself.

Nest-o-patch 2.1.9

New feature: In the Analysis of single channel kinetics, a user can now define arbitrary width and bins number for binned histograms of dwell times in opened / closed states. Bug fix: Error causing appearance of extra symbols in the names of HEKA file sections in the Pulse Tree was fixed. User interface: 1. Context sensitive help invoked with F1 key is implemented. 2. Main Window opens a file with file name drag-drop from a file manager or windows explorer.

Installation.

Download the installer. Start the installer. When Windows asks if you want to execute this unknown application, answer "Yes". If Windows 10 prevents start of it, select "Additional information" and in the opened window say that you want to execute the application. Later follow the instructions of the installer.

Nest-o-patch 2.1.7. Release candidate.

New feature: Calculation of open probability in large ensembles of channels with noise analysis added. Bug fix: Error when built-in calculator did not accept paste from the Notebook was fixed. User interface: If the Notebook was manually edited, the program would ask for saving before closing.

Nest-o-patch 2.1. Official release.

Small bugs which were found and reported during beta-testing were fixed. Manual which covers all new features and underlying theory was included in the release.

Differences with the version 2.1.1-beta 2

  1. Error, when GHK fit with single channels reported limit of outward conductance as 0, is corrected. True value is now reported.
  2. Duplicated hot keys removed in the main menu.
  3. Numbers with leading and trailing spaces can now be pasted into I-V table.
  4. When I-V table was not empty and I-V table for series was calculated, it caused the "Invalid floating point operation" error. It is fixed now.
  5. I-V plot and results of analysis can now be sent to notebook after manual entry of the data.
  6. "Set Selection numerically" command added.
  7. Minor improvements in interface and phrasing of reports.

Differences with the version 2.1.0 (beta 1)

  1. Error in the Trace drawing which could lead to its "punctated" view was corrected.
  2. Eror in HEKA file opening due to unsupported format (Mac PowerPC) is diagnosed correctly (new ReadPulse.dll and ReadPMaster.dll).
  3. Editing of IV table was improved; local menu added.

Differences with the version 2.03

  1. Most important: I-V plot analysis added. It is possible to construct I-V plot from a pulse protocol in one click or manually edit voltages and currents. If reference and experimental series are defined, difference is automatically calculated. I-V plots may be fitted with linear fit, cubic spline or Goldman-Hodgkin-Katz curve. Conductance and, if not known, intracellular concentration of a charge carrier are calculated.
  2. Long-awaited property: the program can recall previous position of Selection. This is particularly useful for calculation of experimental and reference I-V plots. because allows easy selection of equal intervals.
  3. User can navigate and resize the Trace and I-V Plot diagrams with the mouse wheel. Only wheel moved horizontally, [Shift]+wheel moves vertically, [Ctrl]+Wheel resizes.

Interface changes

  1. Design of Pulse tree Tab was refined.
  2. To keep main toolbar reasonably compact, some buttons were moved from it to local toolbars in the tabs.

Bug fixes and optimisations:

  1. Program will not crash when uncontigous sweep and series are selected for highlight.
  2. Options dialog was not possible to open under certain conditions. Now this is fixed.
  3. "Toggle" voltage protocols are supported (fix in ReadPMaster.dll).
  4. Holding potential and sweep voltages are correctly defined when also when an amplifier other then EPC9 or EPC10 is used and voltage is set in "Amplifier" window rather then in PGF file.
  5. Loading of long traces was made much faster (up to 10 times!).
  6. Drawing of long tracess was also made faster, but not as much as loading.
