By default buttons 4+5 are assigned to the

Yes I think so (my mouse has only 3 buttons and no wheel,
but I believe the only way XFree handles a scroll wheel is
by assigning buttons 4 and 5 to it).
Honestly, I think having 5 magnifier sizes is slight
overkill anyway - personally I'm mostly using the
medium-sized one. So another option would be to assign the
sizes you like most to, say, button 1 or 2:
