It would be better if you could use the buttons stock Quit Cancel rather than Yes No. Using labelled buttons instead of yes no saves users the effort of really needing to read anything more than the buttons.
if you want to be really serious you could even switch PyScrabble to use the Gnome button order of Cancel Quit and then use the Gtk Alternative button order feature to give windows users the layout they are more used to.

sorry, forgot to think of the really big picture and mention that if you were to make sure PyScrabble didn't lose any unsaved information and was able to startup fairly quickly there would be no need for a quit warning. having said that it is easier to just tweak the dialog such as it is but i try to present suggestions to developers and let them decide what approach to take.
