It's used by sys.byteorder :-), although I don't know of that variable is set using information from configure. unicodeobject.c does use WORDS_BIGENDIAN. That's the only place using that specific define. The various SIZEOF macros are used all of the place. If haven't checked if those defines could safely be changed into run- time constructs (WORDS_BIGENDIAN) or "calls" to sizeof().
