You can specify the interface and port to use on the command line
like this:-

TG55Editor -i 'MidiSport 2x2' -p 0 -I 'MidiSport 2x2' -P 0

or connect the ports manually after starting it up.

The switches in lower case specify the output port, and the uppercase
specifies the input port.

Download only downloads from the TG55's edit buffer, so to edit an
arbitrary voice you first need to select that voice (either preset
or internal) and then hit the EDIT/COMPARE key to copy the voice
definition to the edit buffer. Then click download. I use the Front
Panel app to navigate between voices and hit the necessary keys.
I'm just used to working that way (I find it helps me avoid overwriting
voices by accident) but if other users find it too bizarre I might be
persuaded to put some controls on the dialog to select the voice number
and PRE or INT. Let me know.
