DeLux provides a MATLAB software to correct for luminescence bleed-through between neighbouring wells on a microplate and estimate the “true” luminescence activity.

INSTALLATION INSTRUCTIONS

1. Unzip the software file DeLux_1.0.zip

2. Open the extracted DeLux folder
3. From MATLAB, run DeLux_main.m
4. The script provides a functional example. To pass custom files and configurations, please read the DeLux_user_manual.pdf


QUICK START GUIDE

1. If your data files are not in TECAN format, please convert them by using the provided tool data_conversion.m and save the .csv output to .xlxs by using an external software
2. In the folder Configs, modify the configuration file userconfig.m to provide information on the plate reader set-up (background wells, tag names to identify luminescence and OD data)
3. Open Run DeLux_main.m and pass calibration file name(s) via cal_filename = {{'file1.xlsx';'file2.xlsx',...}}
4. Pass the experiment file name(s) via exp_filename = {{'file1.xlsx';'file2.xlsx',...}}
5. Run DeLux_main.m
6. The script generates a deconvolution kernel according to the calibration plate data, corrects the experiment for luminescence bleed-through, produces some plots and saves outputs in .mat and .csv files.
