TLS encryption is currently only supported with the Java/Linux/Mac viewer. With that viewer, you can press F8 and select "Connection info", or press CTRL-ALT-SHIFT-I, and the "Security type" field in the Connection Info dialog will indicate whether TLS or X.509 encryption is enabled.

That same dialog will also indicate whether SSH tunneling is active (if it is, then "Host" should be "localhost" + some random port.) Similarly, on the Windows native viewer, you can select "Connection info" from the System Menu or press CTRL-ALT-SHIFT-I to pop up the Connection Info dialog and look at the host and port to see whether your connection is tunnelled. The Windows native viewer doesn't currently support TLS encryption, but there is an open feature request for that (https://github.com/TurboVNC/turbovnc/issues/8).
