Once I have installed Wiki and it's working, how can I get rid of the intial FrontPage? Obviously this isn't something that I want users to see when the enter my site.

Can I change the default page?
You can edit any page you want. In fact, by default anyone can edit any page.
However, by default PhpWiki goes to "HomePage" (in English-- see index.php for language, and and config.php for HOME_PAGE).
Thus, just edit the page.
