I suppose you want to run it from any machine...

Here is a procedure which works for me.
Install ooRexx "normally" in Program Files, and then do a copy of the ooRexx directory to your USB thumbdrive.
Add the script setenv.bat to your USB ooRexx directory.
Before using ooRexx from the USB, open a cmd shell and call setenv.bat.

      set REXX_HOME=%~dp0%
      set REXX_HOME=%REXX_HOME:~0,-1%

:: The (optional) extension directory contains additional binaries
:: like curses, gci, rexxtrans, rexxml, rxcurses
      set PATH=%REXX_HOME%;%REXX_HOME%\extensions;%PATH%

      set PATHEXT=%PATHEXT%;.REX
      set PATHEXT=%PATHEXT%;.ORX
      set PATHEXT=%PATHEXT%;.RXS

      set BSF4REXX_HOME=%REXX_HOME%\extensions\bsf4rexx
      set PATH=%BSF4REXX_HOME%;%PATH%
      set CLASSPATH=%BSF4REXX_HOME%\bsf-rexx-engine.jar;%BSF4REXX_HOME%\bsf-v244-20080704.jar;.;%CLASSPATH%;

:: No space before the redirection
      ftype oorexxfile="%REXX_HOME%\rexx.exe" "%%1" %%> nul
      ftype ObjectRexxScriptFile=%SystemRoot%\System32\WScript.exe "%%1" %%
      > nul

:: No space before the redirection
      assoc .rex=oorexxfile> nul
      assoc .orx=oorexxfile> nul
      assoc .rxs=ObjectRexxScriptFile> nul

      regsvr32 /s orxscrpt.dll
