You are diving in to the lowest level without thinking about the bigger
picture.  The problem is not inside libusb.  Show us YOUR source code.

What kind of device are you driving?  Is this a device you designed? 
Have you verified that there is a bulk pipe with the endpoint number you
are using?  Are you reading or writing?  Are you sure you are reading
from an IN pipe, or writing to an OUT pipe?
