So, I don't really got what these options are for, can someone explain better please?

Upmix Content: If an application plays back stereo or 5.1 sound, then you can upmix it here to 7.1, as long as your device is also set up to 7.1 or stereo.
Content Format: A program might only run internally with 2 or 6 channels and fill up the rest with silent channels, thus you can handle it here like a stereo or 5.1 input.

Automatic means it will use whatever the application uses. Only change this setting if you are sure that you want everything to be stereo + upmix (or 5.1 + upmix). For upmixing this is ideally the same as setting your device back to 5.1 or stereo in Windows.

Thanks, but I still dont know what "upmix" means or how do I know if an game/program runs on stereo or 5.1 by default, or how do I configure "on the OS level". Or why I can select stereo AND 5.1 on the upmix inside HeSuVi (or even disable both).

And in the Content Format I don't know which is the best option.

Best to just leave everything at default then. Your OS audio format is what audio channel format you configured for your device inside the sound panel of Windows.
