I don't use windows so I can't help too much. But it seems strange
that you extracted the contents to python22/gnuplot and then ran
setup.py--normally you would do one or the other:
1. Extract it to python22/Gnuplot then it should be ready to run, or
2. Extract it elsewhere and then run setup.py which installs it to
python22/Gnuplot.
Method 2 is generally recommended. Maybe this helps?
