Mitra, I hope you mean DEFAULT_LANG_FILE. This setting is used with mini-gettext much like it was before with language constants. There is no problem with it. There is only one new $XMGETTEXT_DESTINATION_DIR variable in config.php3, which is important only when you update language files, i.e. not all superadmins must set it.
