Seems like UltraStar does not even start on Windows Vista. Could not test it myself since I do not have Vista.

Right, doesn't fully work on my Laptop with Vista either (Acer Aspire 5920G). While playing the game, it (sometimes) hangs every few seconds and therefore, the audio and the lines get asynchron (the lines are faster). I've looked at the CPU-usage, it's always very high at most songs for the one core(although the Core2Duo T7300 should be fast enough). Compatibility modes didn't fix this problem either.

For me it started fine, but I cannot get the microphone to work even though the same USB microphone works fine with Skype etc

Try to run with mic put to jack input port in your soundcard :) a problem isnt with vista but wit some of da audio who recognises mic input only when a mic is inserted. Ultrastar couldnot work without audio input so tere's a crash
