First of all, the standard disclaimer: using Amazon in the library environment may not be in keeping with Amazon's terms of service (which are pretty explicit about driving traffic to Amazon's site). VuFind supports it because the feature was added long ago when terms were different, and I hate to remove a feature that works -- but use at your own risk.
That being said, the easiest way to compare Amazon vs. Syndetics covers would be to comment out one source, clear your image cache in /stage/local/vufind/web/images/covers/small (also medium/large directories if you want to be thorough), and then look at results... then clear the cache, switch the configuration to the other source, and refresh to see what changes. There isn't currently a more scientific way to keep track of hits at once service vs. another, though it wouldn't be too hard to edit bookcover.php to add some logging capabilities for analysis.
Currently, bookcover.php only really accounts for ISBNs -- the values are passed in from the Smarty templates as an ?isn= parameter. To add support for other types of data, you would need to make sure the necessary details are loaded in the index, that your record driver offers a way of making the data available to the templates, that the templates pass the data to bookcover.php as new GET parameters, and that bookcover.php itself is expanded to process those incoming parameters. A lot of steps, but hopefully not too difficult (the hardest part is probably making sure that every template referring to bookcover.php is appropriately modified -- there are a few different places that display covers).
